Analysis genetic biodiversity with deep sequencing, from single genome to environmental genomics.

Investigating biodiversity implies traditionally tools for collecting samples from environment and rules to examine and to compare biological traits. In genetics, for decades the cost to acquire DNA sequences led to consider only single individuals from a few species as genomic models. With recent progress of sequencing technologies, genomic sciences are being revolutionized due to the possible accessibility of fractions of population at the genomic level and also at the transcriptional level. Also, the time has come to work on groups of species that remain poorly studied though their ecological crucial role such as plankton communities. In this context, we participate to Tara-Ocean, a multi disciplinary scientific project based on a circum-global expedition on all oceans over 3 years. We are assessing the impact of New Generation Sequencing on the feasibility to analyse at the genomic and at the transcriptomic level the diversity of marine protists. The first phase of the project is to establish a set of methodologies enabling the assessment of the real complexity of these populations, and to constitute a significant collection of individual genome sequences from diverse phylums. We present the strategy, preliminary results indicating the relative impact of the different sequencing technologies on a survey of different cell sizes of Mediterranean plankton communities at different depths.
